By default, the Exchange 2010 built-in anti-spam is not installed / enabled and, to be honest, is not a very used solution as it is not very manageable and does not offer great options. In any case if you want to use it you should enable it via a script in the exchange scripts folder, whose default path is:
C:program filesmicrosoftexchangeV14script
within that path there is the script to be launched via powershell:
.install-AntispamAgents.ps1
once launched you must restart the Transport service and then execute this other command to specify the SMTP servers of your exchange infrastructure (if there is only one exchange server that is the only one you need to configure):
Set-TransportConfig -InternalSMTPServers 10.0.1.10,10.0.1.11
where the ips are exactly those of the servers with the HUB role. At the end you need to close and reopen the EMC and you will find a new tab to manage the antispam section in Organization->Hub
